毒邪是一类重要的致病因素，既可由外侵入，也可由内产生，与多种疾病的发生和演变密切相关。深入研究毒的概念、内涵，从源头上剖析其致病特点，对一些重大疑难疾病的防治具有重要的意义。历史上，尤其是清代重视毒的研究，曾对深化认识外感温病，提高温病的防治效果起到了重要作用。当前随着社会的演变、环境污染的影响和饮食结构的改变等，使疾病谱也发生了变化。如何深入研究目前一些重大疑难疾病，搞清其发病机制并提高其临床疗效，成为医务工作者关注的焦点。为此，学者常富业等对于现代疑难性疾病和增龄衰老性疾病的研究过程中，逐渐认识到毒于其中的作用。相继提出了毒邪入络、损络等系列学说，掀起了国内毒邪研究的高潮。为深入研究毒，诠释毒的概念和内涵，剖析毒的致病特点和临床表征，弄清其致病靶位和易损环节，进而确定相应的干预方案，在此其就《黄帝内经》中关于毒的经典论述进行了初步梳理、诠释。
《黄帝内经》中对毒的论述，主要集中于《素问》部分，出现于12篇共34处。大致对毒的认识，主要有以下几个方面。
1病因之毒：此所称的病因之毒，主要指六淫之毒或外毒，但也暗示有内涵更深邃的“毒”。如在《素问·生气通天论》论之毒当属于六淫之毒或外毒，“故风者，百病之始也，清静则肉腠闭拒，虽有大风苛毒，弗之能害，此因时之序也。”于《素问·五常政大论》的前半部分所论之毒，既属于药物之毒的范畴，也属于病因学的范畴之毒，且有更多的内涵。 指出“寒热燥湿，不同其化也。故少阳在泉，寒毒不生，其味辛，其治苦酸，其谷苍丹。阳明在泉，湿毒不生，其味酸，其气湿，其治辛苦甘，其谷丹素。太阳在泉，热毒不生，其味苦，其治淡咸，其谷黅秬。 厥阴在泉，清毒不生，其味甘，其治酸苦，其谷苍赤，其气专，其味正。少阴在泉，寒毒不生，其味辛，其治辛苦甘，其谷白丹。太阴在泉，燥毒不生，其味咸，其气热，其治甘咸，其谷黅秬。化淳则咸守，气专则辛化而俱治。”文中提出了“寒毒”“湿毒”“热毒”“清毒”“燥毒”等名词，这些毒，显然指病因，为外邪所演化而来，提示毒不是独立的一种致病因素，而是邪气演变的产物。按照天人相应的观点，人之五脏六腑犹如天地之五运六气，天地运化能生外寒湿热燥，人之变化亦能生内寒湿热燥。外寒湿热燥能演变为“寒毒”“湿毒”“热毒”“燥毒”，内寒湿热燥亦能演变为“寒毒”“湿毒”“热毒”“燥毒”。以上对毒的提纲论述，为后人认识毒指明了方向。
